Serum Metabolic Fingerprints Characterize Systemic Lupus Erythematosus

Shunxiang Li,
Huihua Ding,
Ziheng Qi
et al.

Abstract: Metabolic fingerprints in serum characterize diverse diseases for diagnostics and biomarker discovery. The identification of systemic lupus erythematosus (SLE) by serum metabolic fingerprints (SMFs) will facilitate precision medicine in SLE in an early and designed manner. Here, a discovery cohort of 731 individuals including 357 SLE patients and 374 healthy controls (HCs), and a validation cohort of 184 individuals (SLE/HC, 91/93) are constructed. Each SMF is directly recorded by nano‐assisted laser desorptio… Show more

“…The high number of carboxylic ions on CNC chains afford strong potential interactions with metal nanoparticles, and thus, AuNPs can be stable for several months in reaction systems. [ 19 ] These results indicated that CNC facilitated high and stable AuNP loading onto CNC. In this reaction mixture, the HAuCL 4 concentration and pH are pivotal in stabilizing AuNPs/CNC.…”

“…These nanomaterials have high absorption coefficients in the ultraviolet (UV)-vis range, which are ideal candidates for supporting LDI, such as gold nanoparticles (AuNPs), carbon nanomaterials, and porous silicon. [14][15][16][17][18][19][20][21] Nanomaterials are mainly used as matrices to analyze small molecules because these materials provide a source of ionization that ensures minimal interference in the low mass range. [22][23][24][25][26][27][28][29] Prompted by rapid progress in techniques, nanomaterials in different derivatized forms are also used as LDI matrices for analyzing peptides and proteins.…”

“…In detail, we initially matched accurate mass and secondary mass spectra within LC‐MS/MS with the human metabolome database (https://hmdb.ca) for metabolite recognition. [ 33 ] Then, the fold change in signal intensity between the two groups was also taken as an important reference for metabolite identification. Particularly, the LC‐MS/MS results were consistent with the LDI MS results, further enhancing the reliability of biomarkers.…”

“…These four putative biomarkers did not correlate with SLEDAI, implying that they are targeted toward SLE diagnosis, however, the ability to assess biomarkers is limited. (Li et al, 2023).…”
