Sesamin attenuates PM2.5-induced cardiovascular injury by inhibiting ferroptosis in rats

Abstract: The present study provided the first in vivo evidence that Ses pretreatment potentially represents a novel and pragmatic therapeutic strategy to protect the cardiovascular system from damage via its anti-ferroptosis activity.

“…Several studies have reported that Ses exhibits its antiinflammatory and antioxidant effects on cardiovascular diseases and metabolic disease through multiple signaling pathways. 15,30 Fan et al 31 reported that treatment with Ses could mitigate myocardial-infarction-induced cardiac damage by reducing cardiomyocyte apoptosis and inflammatory response. 31 With the deepening of research, the protective effect of Ses on the lung has also been gradually discovered.…”

“…Some natural compounds extracted from plants have multiple targets, which can simultaneously modulate multiple signal transduction pathways and exert pharmacological effects. ,, Ses, a natural polyphenolic compound exacted from sesame seeds, exhibits strong anti-inflammatory and antioxidative effects. “…Yoshida et al (2019) reported that accumulation of unstable iron and enhanced lipid peroxidation occurred in cells exposed to CS, and that the accumulation of iron was more severe after GPX4 was knocked out, indicating that ferroptosis may participate in the process of COPD. Furthermore, particle matter 2.5 (PM 2.5) has been shown to induce ferroptosis; the mechanism may be closely related to iron overload, lipid peroxidation, and redox imbalance (Ren et al, 2021).…”

“…Acute lung injury is characterized by acute systemic inflammation. Its clinical manifestations include pulmonary edema, hypoxemia, diffuse alveolar injury, and lung infiltration (Ren et al, 2021). Acute lung injury can further cause multiple organ dysfunction syndrome or acute respiratory distress syndrome.…”

“…In addition, oral administration of 50 mg/kg of sesame lignans attenuated oxidized LDL in rabbits fed a fat- and cholesterol-enriched diet and reduced inflammatory lesions in liver induced by a high-fat diet [ 90 ]. Among the sesame lignans, Ses also alleviated cardiovascular injury in rats by reducing secretion of lactate dehydrogenase (LDH), creatine kinase (CK), and CK-MB in the heart and serum [ 91 ]. In addition, Ses reduced iron accumulation in the heart by recovering the proteins related to ferroptosis inhibition including solute carrier family 7 member 11 (SLC7A11) and glutathione peroxidase 4 (GPX4).…”
