SETD7-mediated monomethylation is enriched on soluble Tau in Alzheimer’s disease

Abstract: Background Human tauopathies including Alzheimer’s disease (AD) are characterized by alterations in the post-translational modification (PTM) pattern of Tau, which parallel the formation of insoluble Tau aggregates, neuronal dysfunction and degeneration. While PTMs on aggregated Tau have been studied in detail, much less is known about the modification patterns of soluble Tau. Furthermore, PTMs other than phosphorylation have only come into focus recently and are still understudied. Soluble Tau… Show more

“…These findings align with a recent 2-year retrospective cohort study that reported an increased risk of psychotic disorder, cognitive deficit, dementia, and epilepsy or seizures persisted in long COVID patients 41 . Furthermore, downregulation of Ddit4, Slc38a2, Tmem267m, Lrrc8c, and setd7 expression levels in the brain at 4 months pi suggests ataxia, impairment of memory, synaptic plasticity, motor, and cognitive abilities, neuronal dysfunction and degeneration in the surviving mice [37][38][39][40] . RNA-seq analysis revealed activation of immune pathways in the CNS, including "complement activation pathway," "phagocytosis recognition," and "humoral immune response mediated by circulating immunoglobulin" at 1 month and 4 months pi.…”

“…Q-PCR analysis was next utilized to determine 8 differentially expressed genes identified by transcriptomic analysis. Reduced levels of expression of Ddit4, Slc38a2, Tmem267m, Lrrc8c, and setd7 genes were noted at 4-month pi, which are associated with ataxia telangiectasia neurodegenerative disease, impairment of memory, synaptic plasticity, motor, and cognitive abilities, neuronal dysfunction and degeneration, and cerebral ischemic stroke respectively [37][38][39][40] (Fig. 6B).…”

“…Furthermore, SYP and TLR4 were found to be the targets of miR-374b-5p and miR-15b-3p, respectively. Notably, SETD7, functioning as a lysine methyltransferase, has implications in the posttranslational modification of tau protein, influencing its subcellular localization (33). SV2B plays a crucial role in regulating neurotransmitter release, which is widely dispersed everywhere in brain.…”

“…Sci. 2023, 24, x FOR PEER REVIEW 2 of 10 methyltransferases have been identified except for SETD7 lysine methyltransferase [10,11]. Tau methylation patterns are significantly different between cognitively normal and AD brains based on mass spectrometry studies [9,12,13].…”

“…Of these PTMs, tau methylation has been less extensively studied, and much is unknown about its effects on tau structures and functions [10]. Very few tau methyltransferases have been identified except for SETD7 lysine methyltransferase [10,11]. Tau 2 of 10 methylation patterns are significantly different between cognitively normal and AD brains based on mass spectrometry studies [9,12,13].…”
