Sex Differences in Blood HDL‐c, the Total Cholesterol/HDL‐c Ratio, and Palmitoleic Acid are Not Associated with Variants in Common Candidate Genes

Abstract: Blood lipids are associated with cardiovascular disease (CVD) risk. Moreover, circulating lipid and fatty acid levels vary between men and women, and evidence demonstrates these traits may be influenced by single nucleotide polymorphisms (SNP). Sex-genotype interactions related to blood lipids and fatty acids have been poorly investigated and may help elucidate sex differences in CVD risk. The goal of this study was to investigate if the influence of SNPs previously associated with blood lipids and fatty acids… Show more

“…In reproductive age, it is known that women and females mice show higher HDL cholesterol level than men and male mice, respectively (Klingel et al, 2017;Link et al, 2015;Wang et al, 2011). These sexual differences could be related with the fact that females have two X chromosomes, which increased the expression of genes escaping X-inactivation (Link et al, 2015).…”

“…DNA was extracted from whole blood for all participants using the Qiagen Paxgene Blood DNA kit (Qiagen, Toronto, Canada) according to manufacturer's instructions, and genotyping was performed using the Sequenom MassArray platform, as previously reported (Klingel et al, 2017). The genotypes for SNP rs174537 for FADS1 and rs953413 and rs9393903 for ELOVL2 were identified for all 86 participants.…”

“…DNA was extracted from either venous blood ( n = 90) or saliva ( n = 110), as previously described (Klingel et al . ). Briefly, 2 mL of saliva was collected in an Oragene DNA collection kit and DNA was extracted according to the manufacturer's instructions (DNA Genotek, Ottawa, ON, Canada).…”

“…All SNPs had a minor allele frequency (MAF) >10% in a European population according to dbSNP (https://www.ncbi.nlm.nih.gov/projects/SNP/). DNA was extracted from either venous blood (n = 90) or saliva (n = 110), as previously described (Klingel et al 2017). Briefly, 2 mL of saliva was collected in an Oragene DNA collection kit and DNA was extracted according to the manufacturer's instructions (DNA Genotek, Ottawa, ON, Canada).…”
