SGT1 is essential for Nod1 activation

Abstract: The Nod-like receptor family in man contains proteins that recognize invasive bacteria. Nod1, a member of this family, is activated by specific peptidoglycan-derived muropeptides that contain meso-diaminopimelic acid. Plants contain a large family of proteins known as resistance (R) proteins that have common structural features with the Nod-like receptors and are essential for protection against a variety of plant pathogens. Extensive genetic studies have shown that the R protein function is determined by mult… Show more

“…NOD2 also sensitizes epithelial cells to recognize H. pylori in vitro (19). NOD1 activity is regulated by its interacting proteins such as cIAP1/2 (26), SGT1 (27), CENTB (28), and Hsp90 (29). Multiple proteins, including TAK1 (30), , Ipaf-1 (31), Erbin (32), and CENTB1 (28) interact with NOD2 and regulate NOD2-mediated NF-κB activation.…”

“…Unlike bE119G, none of the DN mutants in SGSb was able to confer recovery of Rx protein accumulation to wild-type levels (see Supplemental Figure 7 online). The SGS domain is predicted to be the interaction domain for certain LRR proteins, including NBS-LRR proteins Mla1 from barley and human Nod1 (Dubacq et al, 2002;Bieri et al, 2004;da Silva Correia et al, 2007). Thus, we are currently investigating how these SGS mutations affect the formation and/or activation of R protein complexes.…”

“…In addition to structural similarities, NLRs and NB-LRRs have shared function and their stability is regulated by a chaperone complex containing HSP90 and SGT1. [11][12][13] Both NB-LRRs in plants and NLRs in mammals are classified according to their N-terminal domain architecture. Two main classes of plant NB-LRRs have been described: CC-NB-LRRs contain a predicted coiled-coil N-terminal domain and TIR-NB-LRRs carry N-terminal homology to the intracellular TIR domain of TLRs.…”
