Shear Speed-Regulated Properties of Long-Acting Docetaxel Control Release Poly (Lactic-Co-Glycolic Acid) Microspheres

Abstract: Advanced drug carriers for the controlled release of chemotherapeutics in the treatment of malignant tumors have drawn significant notice in recent years. In the current study, microspheres (MPs) loaded with docetaxel (DTX) were prepared using polylactic-coglycolic acid copolymer (PLGA). The double emulsion solvent evaporation method is simple to perform, and results in high encapsulation efficiency. Electron micrographs of the MPs showed that controlling the shear rate can effectively control the size of the … Show more

“…In a different study, Zheng and co‐workers used poly lactic‐ co ‐glycolic acid copolymer to prepare microspheres loaded with docetaxel by using a double emulsion solvent evaporation approach and reported high encapsulation efficiency. [ 140 ] The researchers highlighted that their innovative formulation exhibited a regulated degradation timeline characterized by an initial rapid release, followed by a gradual decrease, ultimately leading to a sustained release until completion. The initial burst release of docetaxel enabled the rapid attainment of an effective drug concentration, after which the release was sustained.…”

“…Additionally, it is difficult to generate homogeneous PLGA particulates using the emulsion solvent evaporation technique. The processing parameters of this technique, such as shearing rate, and stirring rate require stringent control; otherwise, they will produce PLGA particles with varying sizes [ 76 ]. Progesterone-loaded PLGA microparticles prepared by emulsion solvent evaporation and electrospraying methods are different in size, with the electrospraying method generating smaller particle sizes under a high voltage electrical field [ 27 , 77 ].…”
