Short Report: Detection of 72–75-kD and 123-kD Fractions of Leishmania Antigen in Urine of Patients with Visceral Leishmaniasis

Abstract: Abstract. Two polypeptide fractions of 72-75 kD were detected in the urine of 14 of 15 patients with visceral leishmaniasis (VL) and another fraction of 123 kD was found in 10 of the 15 patients by using a Western blot technique. None of these fractions was detected in the urine of 20 controls. These results suggest that antigen detection in urine could be a powerful, noninvasive method for VL diagnosis.

“…Therefore, we cannot discard that some of the bands found in our electrophoretic study can be an antigenic protein of the parasite. However, none of the two antigenic proteins described by De Colmenares et al [6] seems to be equivalent to the bands detected in this study. First, we did not detect any band with a molecular weight around 123 kDa that could be equivalent to the higher molecular weight band previously published [6].…”

“…For this the simultaneous occurrence of two heat shock proteins Hsp70 and Hsp83 (Skeiky et al 1997). In another study, two polypeptide fractions of 72-75 kDa were detected in the urine of 14 of 15 patients with visceral leishmaniasis (VL) and another fraction of 123 kDa was found in 10 of the 15 patients with 96 % sensitivity and 100 % specificity (De Colmenares et al 1995).…”

“…g) Detection of72-75 kDa and 123 kDa fractions of Leishmania antigen in urine may be used to diagnose kala azar [9]. These tests are particularly useful for diagnosis in subclinical infections and in population screening as they are more sensitive and specific than the conventional aldehyde test.…”
