Significant and Conflicting Correlation of IL-9 With Prevotella and Bacteroides in Human Colorectal Cancer

Abstract: Background and aimGut microbiota (GM) can support colorectal cancer (CRC) progression by modulating immune responses through the production of both immunostimulatory and/or immunosuppressive cytokines. The role of IL-9 is paradigmatic because it can either promote tumor progression in hematological malignancies or inhibit tumorigenesis in solid cancers. Therefore, we investigate the microbiota–immunity axis in healthy and tumor mucosa, focusing on the correlation between cytokine profile and GM signature.Metho… Show more

“…This finding may corroborate the existence of a possible link between CA II dysregulation and the immune system in cancer 24 . Despite this evidence, none investigated the expression of CAs in tumour-infiltrating lymphocytes (TILs) that, according to the immunosurveillance theory, were suggested to have a crucial role in developing tumours, as we have previously documented in human CRC 25 , 26 . In addition, growing evidence suggests that tumour mutation burden and TILs are linked to ICI response 27–29 .…”

“…However, IFN-γ can promote CRC proliferation and metastasis by inducing a proinflammatory effect [44], which is in agreement with our findings. In addition, IL9 expression in CRC samples has been reported to be significantly higher than that in the surrounding healthy mucosa, suggesting its tumorigenic role in CRC [45].…”

“…It promotes CRC by the induction of epithelial cell proliferation [ 24 ], enabling a proinflammatory microenvironment [ 25 ] and producing proteins able to stop the antitumoral activity of T and NK cells [ 26 , 27 ]. The data of our recent study [ 28 ] suggest that microbial communities can drive and modulate the antitumor immune response. We have shown for the first time that in CRC, Prevotell a and Bacteroides species are correlated positively and negatively, respectively, with the secretion of IL-9, which has a fascinating and still debated role in tumor immunity.…”
