Significantly reduced lymphadenopathy, salivary gland infiltrates and proteinuria in MRL-lpr/lprmice treated with ultrasoluble curcumin/turmeric: increased survival with curcumin treatment

Abstract: ObjectivesCommercial curcumin (CU), derived from food spice turmeric (TU), has been widely studied as a potential therapeutic for a variety of oncological and inflammatory conditions. Lack of solubility/bioavailability has hindered curcumin's therapeutic efficacy in human diseases. We have solubilised curcumin in water applying heat/pressure, obtaining up to 35-fold increase in solubility (ultrasoluble curcumin (UsC)). We hypothesised that UsC or ultrasoluble turmeric (UsT) will ameliorate systemic lupus eryth… Show more

“…reported enhancement of the solubility of curcumin in water by 35 folds by heat under pressure. Interestingly, heat‐solubilized curcumin was shown to increase pharmacological effect of curcumin in mice with systemic lupus erythematosus (SLE) and Sjögren's syndrome (SS) . This finding is in line with previous studies showing enhanced solubility and biological activity of heat‐treated curcumin .…”

“…Mice were killed at day 7, 14 and 21 [36]. For the lupus model, groups of female MRL/WT or MRL/lpr mice with spontaneous lupus-like autoimmunity were killed at 6, 10 and 14 weeks as described [37,38]. MRL/lpr female mice develop a lupus-like disease at week 10, which is further aggravated in the next weeks.…”

“…Thus, week 14, a time point when the mice already show fully developed the lupus-like disease, was chosen. Week 6 was used as a baseline control [38]. Kidneys and spleens were harvested for RNA isolation and RT-PCR.…”

“…(b) heat-solubilized curcumin inhibited 4-hydroxy-2-nonenal oxidation of a protein substrate by 80%,[19] supporting the reported role of curcumin as an antioxidant,[456] (c) heat-solubilized curcumin inhibited antibody-antigen interaction,[20] (d) heat-solubilized curcumin could serve as a sensitive protein stain in SDS-PAGE gels,[21] (e) heat/pressure (121°C)[22] solubilized curcumin ameliorated lupus like disease in mice.…”
