Silencing acetyl-CoA carboxylase A and sterol regulatory element-binding protein 1 genes through RNAi reduce serum and egg cholesterol in chicken

Abstract: Cholesterol is synthesized in chicken through de novo lipid biosynthetic pathway where two most important genes viz. SREBP1 and ACACA play immense role. To minimize cholesterol synthesis, RNAi approach was adopted and accordingly, we developed transgenic chicken possessing ACACA and SREBP1 shRNA constructs, which showed lower level of ACACA and SREBP1 in serum. The serum total cholesterol, triglycerides, HDL and LDL cholesterol was significantly lower by 23.8, 35.6, 26.6 and 20.9%, respectively in SREBP1 trans… Show more

“…However, HDL levels decreased in HC chicks, while TG levels exhibited a 5-fold increase compared to the ST group. These changes in lipid metabolism are likely the result of a metabolic shift induced by earlier access to feed, as corroborated by findings of other researchers regarding changes in the expression of genes responsible for lipid metabolism and absorption in hepatic cells and intestinal epithelial cells ( Tsunekage and Ricklefs, 2015 ; Metzler-Zebeli et al, 2019 ; Zaytsoff et al, 2019 ; Prasad et al, 2022 ).…”

“…This enzyme is closely related to fat deposition, metabolism, and growth performance in broilers. Mutations or variations in the ACACA gene can affect the fat content [5]: [6]. while The Gpx1 gene, known as Glutathione Peroxidase 1 gene, is a gene present in broiler chickens.…”
