Silibinin-Induced Apoptosis in MCF7 and T47D Human Breast Carcinoma Cells Involves Caspase-8 Activation and Mitochondrial Pathway

Abstract: Silibinin, a natural flavonoid, under phase I/II clinical trial in prostate cancer patients was aimed to evaluate its chemotherapeutic potential in human breast cancer cell MCF7 and T47D. Results showed that T47D cells were found to be more sensitive to silibinin than MCF7 as observed by proliferation, clonogenic, and apoptotic assays, which was abrogated by pan-caspase inhibitor but remained unaffected by p53 inhibitor. Apoptotic events in both cell types differ temporally and also by magnitude that involved … Show more

“…This indicates that other caspase mechanisms are involved in silibinininduced MCF7 cell death. These results are similar to reports that silibinin induces apoptosis through a mechanism independent of caspase-8 activation in MCF7 and T47D breast cancer cells (Tiwari et al, 2011).…”

“…In a transgenic adenocarcinoma of the mouse prostate (TRAMP) mouse model, silibinin inhibits tumor growth, progression, local invasion and distant metastasis [56]. Silibinin induces both death receptor-mediated and mitochondrial-mediated apoptosis in human breast cancer MCF-7 cells [58]. Silibinin also reduces hepatocellular carcinoma xenograft growth through the inhibition of cell proliferation, cell cycle progression, as well as phosphatase and tensin homolog/P-Akt and extracellular signal-regulated kinase (ERK) signaling.…”
