Silver nanoparticles improve structural stability and biocompatibility of decellularized porcine liver

Abstract: No ideal cross-linking agent has been identified for decellularized livers (DLs) yet. In this study, we evaluated structural improvements and biocompatibility of porcine DLs after cross-linking with silver nanoparticles (AgNPs). Porcine liver slices were decellularized and then loaded with AgNPs (100 nm) after optimization of the highest non-toxic concentration (5 µg/mL) using Human hepatocellular carcinoma (HepG2) and EAhy926 human endothelial cell lines. The cross-linking effect of AgNPs was evaluated and co… Show more

“…AgNPs have also been shown to interact with the ECM [ 5 ] and may stabilize the ECM structures in scaffold constructs [ 29 ]. AgNPs are known to regulate collagen deposition, and AgNP composite nanofibers have been shown to increase collagen production during wound healing [ 30 , 31 ] and may thereby indirectly induce tissue remodeling.…”

“…Both the strains were then treated with different concentrations of biosynthesized AgNPs (0, 25, 50 and 100 mg/ml) under optimum conditions of bacterial growth (37 C and 150 rpm continuous shaking). Growth of both the strains in LB liquid medium was measured by optical density (OD) @ k ¼ 600 nm at regular intervals (1,2,4,6,8,10,12,14,16,18, 20, 22 and 24 h) using UV-Vis spectrophotometer. The growth curve was obtained by plotting optical density against time [18].…”

“…Recently, numerous important technological innovations occurred in the field of nanotechnology. Nanoscience and nanotechnology deal with the synthesis of nanoparticles (NPs) of different chemical compositions [1]. At present, the importance of this field emerges in environmental science because of wide applications of nanoparticle for removal of toxic chemicals and pollutants from environment [2].…”

“…Many authors reported that the hepatotoxic effect of the TAA injection depends on ROS release and antioxidant enzyme depletion (Staňková et al, 2010) that, in turn, result in excessive lipid peroxidation and mitochondrial damage, leading to cell death (Koblihová et al, 2014). AgNPs could increase Ki67 expression as they have a role in parenchymal and non-parenchymal cell proliferation (Saleh et al, 2018) which in turn increase HGF secretion. However, in LECM hydrogel group, low HGF and Ki67 expressions were detected.…”

“…The TAA-induced hepatic cell injury in vitro was induced by adding TAA (40 mmol/L) to precultured HepG2 cells for 24 hr in a culturing media (Staňková et al, 2010). After 1 day, TAA containing media was changed by the conditioned media prepared as described elsewhere (Saleh et al, 2018)…”
