Similar coronary vascular effects in the rat perfused heart of platelet‐activating factor structural analogues with agonist and antagonist properties

Abstract: 1 Selective blockade of platelet-activating factor (PAF) receptor subtypes by PAF receptor antagonists has been demonstrated. However, selective activation of PAF receptor subtypes by PAF receptor agonists has not been reported. 2 When structural analogues of PAF that have been shown to possess either agonist or antagonist effects were administered by a bolus injection in the rat perfused heart, they all showed agonist effects. Lower amounts produced vasodilatation while higher amounts produced vasodilatation … Show more

“…In rats, PAFR antagonists reduced infarct size and the occurrence of arrhythmias. 62–64 Although some conflicting results were obtained with different PAFR antagonists, a comparable protective effect was also observed in the dog. 65,66 To date, no clinical trials have investigated the protective effect of PAFR antagonists in cardiac alterations associated with I/R in humans.…”

“…In the experimental myocardial infarction by coronary occlusion, PAF receptor antagonists reduced the hematological and hemodynamic alterations as well as the size of necrotic area and the accumulation of platelets and leukocytes observed in rabbit (287,288) and sheep (213) hearts but did not affect plasma protein leakage (91,287,459). In rats, PAF antagonists reduced infarct size and arrhythmias (144,158,263,265). In dogs, conflicting results were obtained with different PAF receptor antagonists.…”
