Similarity of therapeutic networks induced by a multi-component herbal remedy, Ukgansan, in neurovascular unit cells

Abstract: the neurovascular unit, which includes neurons, glial cells, and vascular cells, plays crucial roles in the onset and progression of Alzheimer's disease (AD). Therefore, effective drugs against AD should be able to target the multi-cellular neurovascular unit and the therapeutic relationships among neurovascular cells should be defined. Here, we examined the therapeutic effects of Ukgansan (UGS), an herbal remedy with multi-targeting capabilities, using in vitro neurovascular unit models and an in vivo model o… Show more

“…Several prior studies used bioinformatic analysis based on omics data, including transcriptome, to address the issue. 31 , 50 , 51 Frankincense contains a wide range of phytochemicals, including volatile oil, polysaccharides, monoterpenes, diterpenes, and lipophilic pentacyclic triterpene acids. 19 , 22 In order to understand the molecular mechanisms behind the pro-apoptotic and anti-adhesive effects of Fr, we also used the transcriptome and bioinformatic analyses in this study.…”

“…The GSEA was carried out using GSEA software from the Broad Institute (version 4.3.0, available at http://www.gsea-msigdb.org/gsea/downloads.jsp ) as described previously. 31 , 32 Analysis of the biological processes of DEGs was done using hallmark, Kyoto Encyclopedia of Genes and Genomes (KEGG), and Gene Ontology (GO). Cytoscape was used to display the results of the network analysis of the GO gene with values of p < 0.001 and q < 0.5.…”

“…The pathway activity was examined by calculating the cumulative effect of the expression of genes included in each pathway [ 30 , 43 ]. In brief, the log-transformed expressional ratios of genes relative to the control in each pathway were added linearly with a weight of −1 for genes that acted as repressors, which were defined as proteins that inhibited the process of signal transduction of the pathway.…”

“…In recent years, research efforts have been directed toward the exploitation of the multi-targeting potentials embedded in herbal medicines in the treatment of diseases with multi-factorial causes [ 23 , 24 ]. These multi-target phytotherapeutic effects are currently being investigated in the treatment of numerous diseases including malaria [ 25 ], Alzheimer’s disease (AD) [ 26 ], heart disease [ 27 ], insulin resistance [ 28 ], T2DM [ 29 ], and NAFLD [ 30 ].…”
