Simple HPLC-UV method for the quantification of metformin in human plasma with one step protein precipitation

Abstract: This study presents the optimization of a simple HPLC-UV method for the determination of metformin in human plasma. Ion pair separation followed by UV detection was performed on deproteinized human plasma samples. The separation was carried out on a Discovery Reversed Phase C-18 column (250 × 4.6 mm, 5 μm) with UV detection at 233 nm. The mobile phase contained 34% acetonitrile and 66% aqueous phase. Aqueous phase contained 10 mM KH2PO4 and 10 mM sodium lauryl sulfate. Aqueous phase pH was adjusted to 5.2. The… Show more

“…Metformin, a biguanide-derivative antidiabetic drug, is the most widely used oral agent in humans (Brinkmann, Brixius, 2015;Chhetri, Thapa, Van Schepdael, 2014). It is the first-line drug for the treatment of type 2 diabetes, particularly, in overweight and obese patients with normal kidney function (American Diabetes, 2009;George, Joseph, 2014;Hung et al, 2015).…”

“…It improves glycemic control by decreasing hepatic glucose production, decreasing glucose absorption and increasing insulin-mediated glucose uptake [7] Various analytical methods have been reported for the estimation of Metformin Hydrochloride as alone as well as in combination with other drugs. They include Titrimetric method [8] , spectrophotometric methods [9][10][11] , HPLC [12][13][14] However an extensive literature search didn't reveal any estimation method for Empagliflozin and Metfromin hydrochloride drugs in their combined dosage form. Therefore, attempt was made to stability indicating method development and Validation of simple, precise, and accurate, RP-HPLC method for simultaneous determination of Empagliflozin and Metfromin hydrochloride drugs in their combined dosage form.…”

“…In general, the use of a photodiode array (PDA) detector provides advantages for sensitive and selective HPLC systems during the analysis of drugs and their metabolites in biological matrices [15,16]. Using these principles, various analytical methods for determining plasma metformin levels have been widely developed and accepted [5,[17][18][19][20].…”
