Simple method for the assay of linezolid in Brain Heart Infusion broth by high‐performance liquid chromatography

Abstract: A simple HPLC method was developed and validated for quantification of linezolid in Brain Heart Infusion (BHI) broth. Eperezolid was employed as internal standard and the sample pre-treatment procedure was simple. Calibration standards ranged from 0.05 to 16 mg/L. Accuracy was within 7.8% and reproducibility (RSD) was less than 8.3%. Recovery was approximately 100% at all concentrations examined. Linezolid was stable in the autosampler insert for at least 24 h at ambient temperature and in BHI for 72 h at 37 d… Show more

“…If extraction recovery reached, ca. 100% after deproteinization of serum [6,9], plasma [8,11], urine [9], microdialysate [8], BHIB [10], it [15]. These last results are close to that obtained for M-HB with our method.…”

“…deproteinization [6][7][8][9][10][11] and solid-phase extraction [12][13][14], were proposed for linezolid determination in plasma, serum, urine, brain heart infusion broth (BHIB) and microdialysis samples. An on-line extraction technique [15] was also applied to serum and urine samples.…”

“…Samples obtained from each treatment arm were stored at Ϫ80°C until assayed for linezolid concentrations. Linezolid concentrations in MHB were diluted with methanol and were analyzed by high-performance liquid chromatography (HPLC) using the method described by Li et al (9). Chromatographic separation was performed using a Kromasil C 18 Ciprofloxacin concentration determinations.…”

“…Samples obtained from each treatment arm were stored at Ϫ80°C until they were assayed for their linezolid concentrations. Linezolid concentrations in MHB were measured by high-performance liquid chromatography using the methods described by Li et al (20). The standard curve ranged from 0.1 mg/liter to 30 mg/liter.…”
