Simplified Method to Isolate Highly Pure Canine Pancreatic Islets

Abstract: Objectives The canine model has been used extensively to improve the human pancreatic islet isolation technique. At the functional level, dog islets show high similarity to human islets and thus can be a helpful tool for islet research. We describe and compare 2 manual isolation methods, M1 (initial) and M2 (modified), and analyze the variables associated with the outcomes, including islet yield, purity, and glucose-stimulated insulin secretion (GSIS). Methods Male mongrel dogs were used in the study. M2 (n … Show more

“…Data on the size of pancreatic islets in healthy dogs are scarce. In a study on different isolation procedures of pancreatic islets in 14 healthy dogs the average dimension of islets varied from 50 to 325 µm, while in 75% of them it was 100 to 200 µm (17). In other studies on healthy beagles and cross-breed dogs the average dimension of pancreatic islets was 50-100 µm (8,10).…”

“…In young dogs (6 months), type B cells are most numerous, representing 52-82% of the total islet cells, and their amount is constant, in contrast to percentages of A and PP cells that differ depending on the region (9). The size of the islets in healthy dogs ranges from 50 to 325 µm (8,10,17).…”

“…Islets and adipose derived MSCs were Isolated and cultured from mice and dogs as previously reported . Purified human islets from nondiabetic subjects (Prodo Laboratories, Irvine, CA), and human adipose derived MSCs (P1, Lonza, Walkersville, MD) were identically cultured.…”

“…The long digestion of the pancreas resulted in islets of high purity although the total number of free islets was low. Most isolated islets had a diameter smaller than 100 μm, which might be associated with islet fragmentation 11 . Considering these findings, enzyme blends of higher purity and more precise composition may be more appropriate to ensure consistent degradation activity without deteriorating islet integrity 3 .…”
