Simplified Rapid Technic for the Extraction and Determination of Serum Cholesterol without Saponification

Abstract: An accurate yet simple procedure for the determination of total cholesterol, based upon the application of a Liebermann-Burchard color reaction directly in the solvent employed for extraction of cholesterol from serum, has been described. Extraction of cholesterol and removal of protein are accomplished by means of acetic acid and acetic anhydride. Serum water is removed by the acid-catalyzed hydrolysis of acetic anhydride. The Liebermann-Burchard color is then developed with a stable, modified reagent consist… Show more

“…The lipid extract was collected in a weighed beaker and dried to constant weight at 60" for total-lipid determination. The lipid extract was redissoIved in the above solvent or cells were saponified and the sterols extracted with petroleum ether for estimation of cholesterol by the Lieberman-Burchard procedure (4). Lipid analyses were done at least 4 times and average results are presented.…”

“…After scanning of the electropherogram in a Photovolt curve resolver with integrator at 650 nm and determination of total serum triglyceride (Laurel1 1966), cholesterol (Carr & Drekter 1956) and phospholipids (Walaas et al 1971), the absolute and relative amounts of the different lipoprotein classes were estimated in accordance with the studies by Dyerberg & HjGrne (1970). In one of the cases the individual phospholipids were determined by thin layer chromatography as previously reported (Walaas et al 1971).…”
