Simultaneous detection of Salmonella spp., Salmonella Typhi and Salmonella Typhimurium in sliced fruits using multiplex PCR

Abstract: a b s t r a c tThis study aimed to determine the prevalence and quantity of Salmonella spp., Salmonella Typhi and Salmonella Typhimurium in sliced fruits from hawker stalls and hypermarkets in Malaysia. Analysis was carried out using the most probable number (MPN) e multiplex polymerase chain reaction (PCR) method. The prevalence of Salmonella spp., Salmonella Typhi and Salmonella Typhimurium in 210 samples of sliced fruits examined were 23.3%, 7.6% and 3.8%, respectively with estimated quantity varying from 0… Show more

“…Salmonella and Campylobacter are the most frequently isolated foodborne pathogens, and are predominantly found in poultry, eggs and dairy products (Silva et al 2011). Other food sources that are involved in the transmission of Salmonella include fresh fruits and vegetables (Pui et al 2011). In general, food animals such as swine, poultry and cattle are the prime sources of Salmonella infections.…”

“…Furthermore, the poultry production systems are designed to ensure the massive production of birds usually in confinement. This design is necessary to meet the high protein demand, it also promote the introduction and establishment of the Salmonella organisms and hence transmission to the consuming population [8,9].…”

“…If only direct PCR was to be performed on the cat/ dog food samples, it is possible for false positive results to occur due to the presence of dead cells. The incorporation of MPN method which allows only viable cells to grow can solve this problem by complementing the PCR method for accurate detection of L. monocytogenes [22]. The primers used in this study amplified a specific region present in the hly gene of L. monocytogenes.…”
