Simultaneous determination of major components of Huangqi–Honghua extract in rat plasma using LC–MS/MS and application to a pharmacokinetic study

Abstract: A sensitive and reliable LC-MS/MS method was developed and validated for simultaneous quantification of the major components of Huangqi-Honghua extact in rat plasma, including hydroxysafflor yellow A (HSYA), astragaloside IV (ASIV), calycosin-calycosin-3′-O-sulfate (C-3′-S). After extraction by protein precipitation with acetonitrile and methanol from plasma, the analytes were separated on a Hypersil BDS C 18 column by gradient elution with acetonitrile and 5 mM ammonium acetate. The detection was carried out … Show more

“…Recently, another group identified as many as 23 original flavonoids constituents from AM in rats after oral administration using the high performance liquid chromatography equipped with photodiode array‐electrospray ionization‐ion trap‐time of flight multistage mass spectrometry technique (Liu, Li, et al, 2020). Furthermore, Wang et al applied an LC–MS/MS method to the pharmacokinetic study of CG and astragaloside IV in rat plasma after an oral administration of Huangqi–Honghua extract (Wang et al, 2019). Zeng et al developed a UPLC–MS/MS method for formononetin, CG, ononin and astragaloside IV to the pharmacokinetic study of multiple components in rat plasma after intragastric administration of Huangqi decoction (Zeng et al, 2018).…”
