Simultaneous Qualitative and Quantitative Evaluation of the Coptidis Rhizoma and Euodiae Fructus Herbal Pair by Using UHPLC-ESI-QTOF-MS and UHPLC-DAD

Abstract: The herbal pair of Coptidis Rhizoma (CR) and Euodiae Fructus (EF) is a classical traditional Chinese medicine formula used for treating gastro-intestinal disorders. In this study, we established a systematic method for chemical profiling and quantification analysis of the major constituents in the CR-EF herbal pair. A method of ultra high performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS) for qualitative analysis was developed. Sixty-five compounds, including alkaloid… Show more

“…Taking compound 18 as an example, it exhibited [M + H] + ion at m/z 370.3099 in the positive ion mode, indicating an MW of 369. In the MS/MS spectrum, a series of characteristic ions— m/z 159.0676 ([C 10 H 9 NO] + ), 173.0838 ([C 10 H 9 NO + CH 2 ] + ), and 186.0918 ([C 10 H 9 NO + C 2 H 3 ] + )—were yielded by the fragmentation of different positions at the side chain, as described in our previous study [ 9 ]. In addition, the fragment ion at m/z 200.1069 was generated by the neutral loss of C 6 H 12 from the parent nucleus.…”

“…In our previous studies [ 9 ], 65 compounds, mainly including alkaloids, phenolic compounds, and limonoids, in the CR–EF herbal pair system were identified by UHPLC–ESI–QTOF-MS analysis, and an optimized UHPLC–DAD method was established for the chemical fingerprint analysis of the CR–EF herbal pair extracts and the quantitative determination of nine major alkaloids. We conducted a comprehensive study in the quality control of CR–EF herbal pairs.…”

“…In addition, binding models of columbamine with MAO-B active sites revealed that it buries itself inside the pocket of MAO-B, a mechanism similar to safinamide inhibition [51]. Through a MAO-B enzyme activity assay, columbamine showed a moderate inhibitory effect by decreasing MAO-B enzyme activity by 40% [52]. As a result, this may be essential in explaining columbamine's partial involvement in nociception within the central nervous system [51].…”

“…It has been identified in various plant species. It is an active component of Coptidis rhizoma, the dried rhizome of Coptis chinensis Franch., C. deltoidea C. Y. Cheng et Hsiao or C. teeta Wall., widely used in Traditional Chinese Medicine [52]. Figure 8 illustrates the chemical structure of columbamine.…”
