Simultaneous quantification of two steroidal glycosides after oral gavage of Marsdenia tenacissima extract in rats using a LC‐MS/MS method

Abstract: A specific, sensitive and accurate analytical LC-MS/MS assay was developed for the simultaneous determination of two steroidal glycosides, tenacissoside H and tenacissoside I, in rat plasma. An Agilent ZORBAX SB-C18 column was used with an isocratic mobile phase system composed of methanol-water-formic acid (70:30:0.1, v/v/v) at a flow rate of 0.3 mL/min. The analysis was performed on a positive ionization electrospray mass spectrometer via selected reaction monitoring mode scan. One-step protein precipitation… Show more

“…Another more sensitive analytical LC-MS/MS method with one-step protein precipitation was proposed to simultaneously determine the plasma concentration of two steroidal glycosides, tenacissoside H, and tenacissoside I, in rat after oral gavage. The lower limits of quantification was 0.9 ng/mL for these two MT extracts ( 66 ). Recently, Yu et al developed a new evaluation model combining DNA barcoding techniques with thin-layer chromatography (TLC) and HPLC.…”

“…So far, several pharmacokinetic studies have been carried out on the steroids of M. tenacissima owing to their clinical applications and significant effects. For example, Wang et al quantified the tenacissoside H and I contents in rat plasma by LC–MS/MS (Wang, Sun et al, 2015). Zeng et al constructed an LC–MS/MS approach for quantifying 17 β ‐tenacigen B, tenacigenin A, tenacigenin B, tenacigenoside A, marsdenoside I and tenacigenoside F in rat plasma (Zeng et al, 2014).…”

“…The derivatives of Tenacigenin B can reverse P‐glycoprotein‐mediated multidrug resistance in HepG2/Dox cells (Hu et al , ) and have cytotoxic activity against cancer cell lines (Luo et al , ). Tenacissoside H and Tenacissoside I are detected at relatively high levels in M. tenacissima , and Tenacissoside H serves as the marker compound for quality control (The State Commission of Chinese Pharmacopoeia, ; Wang et al , ) . Tenacissoside H and Tenacissoside I possess an identical oligosaccharide chain consisting of two sugar units at C‐3 of their aglycone.…”
