Simultaneous Solid Phase Extraction, Derivatization, and Gas Chromatographic Mass Spectrometric Quantification of Thromboxane and Prostacyclin Metabolites, Prostaglandins, and Isoprostanes in Urine

Abstract: The current analytical methods for the various prostanoids require a separate and extended sample workup, derivatization, and gas chromatographic/mass spectrometric detection of each compound. Therefore, we developed and validated a rapid method for the common purification, derivatization, and GC/MS determination of 11-dehydrothromboxane B2, 2,3-dinor-6-keto-PGF1a, PGF2A, PGE2, PGD2, and isoprostanes in urine. A single reversed-phase solid-phase extraction step and modified reaction conditions yielded excellen… Show more

“…Before analysis with gas chromatography (GC), all the samples were dried under a nitrogen stream and derivatized to their trimethylsilylethers. For this, samples were generated for 1 h at 45C in 50 µL of silyl 911 (99% BSTFA + 1% TMCS) according to Wubert et al. (1997).…”

“…Optimal separation of the three analytes was achieved with the following oven program: initial temperature, 150˚C for 1 min, followed by an increase to 280˚C at 8˚C/min, with a final time of 20 min. Specific ions were used for selected ion monitoring (54).…”

“…Their measurement represents one of the most reliable approaches to assess oxidative stress status in vivo [81]. Several methods are used to measure F 2 -isoprostanes: gas chromatography/mass spectrometry [12, 21, 82, 83, 84, 85, 86, 87, 88]; gas chromatography/tandem mass spectrometry [89, 90]or liquid chromatography/tandem mass spectrometry [24]. The main problems with these specific methods are their cost and their technology which limit their routine use.…”
