Simvastatin Induces Apoptosis of Fibroblast-Like Synoviocytes~!2009-02-16~!2009-03-24~!2009-09-07~!

Abstract: Background:Statins (3-Hydroxy-3-methylglutaryl-CoA reductase inhibitors) exert favorable effects on lipoprotein metabolism, but appeared to possess anti-inflammatory properties among others, as suggested by their ability to inhibit collagen-induced arthritis in mice. Their activity in fibroblast-like synovial cells (FLS) has not yet been studied. Objectives:To evaluate the effect of varying doses of simvastatin on apoptosis of FLS.Methods:Synovial tissue, obtained during total knee replacement due to osteoarthr… Show more

“…The 2200-fold upregulation of BNIP3 , a pro-apoptotic protein, in RA FLS led us to investigate apoptosis as a mechanism of observed cell cytotoxicity in TLR2 activated FLS. SMV as a single treatment has previously been shown to cause apoptosis in NHS [32]. Annexin V flow cytometry assay indeed showed a dose dependent apoptosis with SMV (Figure 4 A-D).…”

“…Statins profoundly decrease the expression of osteoarthritic-speciWc matrix metalloproteinases, proteolytic peptidases responsible for tissue remodeling and protein degradation, and induce activated Wbroblast-like synoviocyte apoptosis, eVects that contribute to sustained joint integrity [62][63][64][65].…”
