Single-Cell Metabolomics: Analytical and Biological Perspectives

Abstract: There is currently much interest in broad molecular profiling of single cells; a cell's metabolome-its full complement of small-molecule metabolites-is a direct indicator of phenotypic diversity of single cells and a nearly immediate readout of how cells react to environmental influences. However, the metabolome is very difficult to measure at the single-cell level because of rapid metabolic dynamics, the structural diversity of the molecules, and the inability to amplify or tag small-molecule metabolites. Mea… Show more

“…Another interesting topic is the analysis of metabolic fluxes, also called fluxomics (Mo et al 2009;Celton et al 2012). In budding yeast, the first single-cell metabolomic studies have recently appeared (Ibanez et al 2013;Zenobi 2013), and methods for highthroughput screening of low-volume cultures from 96-well plates are also being developed (Ewald et al 2009 …”

“…There are four major methods used to quantify the metabolome of single cells: Fluorescence-Based Detection, Fluorescence Biosensors, Fluorescence resonance energy transfer (FRET) based-biosensors and Mass Spectroscopy. The first three methods listed use fluorescence microscopy to detect molecules in a cell [9]. Usually these assays use small fluorescent tags attached to molecules of interest.…”

“…At this point, we want to emphasize the many comprehensive review articles that discuss the challenges and current approaches of single cell analysis (e.g., Zenobi [5] Fritzsch et al [6], Schmid et al [7], Borland et al [8]). Furthermore, in a recent issue of ABC (406/14) entitled "Multiplex platforms in diagnostics and bioanalytics," the state of the art of those methods is discussed, which are already fit for purpose of measuring at cellular levels.…”
