Single Nucleotide Polymorphisms in MicroRNA Binding Sites: Implications in Colorectal Cancer

Abstract: Cancer is a complex genetic disorder, characterised by uncontrolled cell proliferation and caused by altered expression of oncogenes and tumour suppressor genes. When cell proliferation pertains to colon, it is called colorectal cancer. Most of colorectal cancer causing genes are potential targets for the miRNA (microRNA) that bind to 3′UTR (untranslated regions) of mRNA and inhibit translation. Mutations occurring in miRNA binding regions can alter the miRNA, mRNA combination, and can alter gene expression dr… Show more

“…MicroRNAs (miRNAs) are endogenous non‐coding small RNAs that regulate a variety of important biological processes by regulating gene expression, including cell development, proliferation, differentiation, apoptosis, signal transduction and tumor occurrence, and development . Computer technology analysis objectively demonstrated the mutation of the miRNA binding site involved in cancer and the mechanism of gene regulation mediated by it . In addition, miRNAs are aberrantly expressed in various types of malignancies and function either as oncogenes or tumor suppressors .…”

“…Most of these studies focus on the correlation of CTC enumeration with prognosis 913. However, recent studies showed that only enumerated CTCs were not enough to reflect the heterogeneous condition of tumors 3,14,15.…”

“…Therefore, the 3′UTR region is emerging as critically important in regulating gene expression, and polymorphisms in the miRNA‐binding sites of the 3′UTR of genes may alter the binding efficiency and miRNA‐mRNA gene expression regulation. In support of this hypothesis, recent studies have identified variants in the 3′UTR of genes that increase the susceptibility to melanoma, lung, colorectal and ovarian cancer by affecting the ability of miRNAs to bind. In particular, two sequence changes in the 3′UTR of the CDKN2A gene have been significantly correlated with melanoma risk, but also with a shorter progression time from primary to metastatic melanoma …”
