SINGLET OXYGEN INVOLVEMENT IN PHOTOHEMOLYSIS SENSITIZED BY MEROCYANINE‐540 and ROSE BENGAL

Abstract: Merocyanine-540 is currently in use in experimental protocols for the treatment of leukemia and neuroblastomas. The mechanism of cellular phototoxicity of this membrane binding sensitizer is unexplored. We have used sodium azide and deuterium oxide to examine the role of singlet oxygen in photomodification of cell membranes in a photohemolysis assay. The effects of these agents on the photomodification process were separated from the effects on the ion fluxes leading to lysis (lytic phase). Azide significantly… Show more

“…RB is a water-soluble dye and a localized anionic group on its molecule inhibits it from penetrating into or through the lipid bilayer. Several studies showed that RB is localized in the plasma membrane of mammalian cells [8,[21][22][23], especially on the outer surface of the membrane [23]. Therefore, we assumed that the site of reaction might be limited in the endothelial cells of vessels and that the cochlear dysfunction might be induced by ischemia, but not by direct damage to the hair cells and/or spiral ganglion cells.…”

“…[4][5][6][7][8][9][10] Second-generation merocyanines have not yet been characterized as thoroughly as MC540. However, all available data are consistent with the notion that when used as agents for PDT, all dyes of Günther's series behave like Type II photosensitizers.…”

“…33 However, the rate decrease observed should not be taken as an argument against a singlet-oxygen mechanism. As discussed by Valenzeno et al 34 the D 2 O effects on photohemolysis using other sensitizers are somewhat variable, ranging from no effect to an enhancement of over 200%. We have shown recently that the binding constant between RBC and InTPP is (2.40 0.05) 10 7 L mol -1 , in aqueous solutions.…”
