Size and Charge Adaptive Clustered Nanoparticles Targeting the Biofilm Microenvironment for Chronic Lung Infection Management

Abstract: Chronic lung infection caused by bacterial biofilms is an extremely serious clinical problem, which can lead to the failure of antibiotic therapy. Although nanoparticles have shown great potential in the treatment of biofilms, the efficient penetration and retention of nanoparticles in biofilms is still a big challenge. To address this issue, we herein fabricate size and charge adaptive azithromycin (AZM)-conjugated clustered nanoparticles (denoted as AZM-DA NPs) as therapeutic agents for treating biofilms. “…glycol)-block-polylysine (PEG-PLys-COOH[DA]) ( Figure 3a). [40] The AZM-DA nanoparticles exhibited a long circulation time due to their suitable size (112.0 nm) and negative surface charge (−2.2 mV) at neutral pH ( Figure 3b). Moreover, they could disassemble in the acidic biofilm microenvironment (pH 6.0) due to the pH sensitivity of the DA moiety, and subsequently released small-sized and positively charged PAMAM-AZM nanoparticles.…”

“…Thus, TK-PPE@NP Ce6/DOX exhibited excellent anticancer performance after laser irradiation (Figure 4c). As stated above, the designed photocontrolled size-shrinkable nanosystems (SiPINGs) could be used to load the anticancer drug DOX, thus [40] Copyright 2020, American Chemical Society.…”

“… Gao et al (2020) showed that azithromycin (AZM) conjugated to PAMAM dendrimer has effective in vitro and in vivo antimicrobial action against Pseudomonas aeruginosa biofilm, which is responsible by chronic lung infection. The use of dendrimer demonstrated many beneficials such as, maintained the AZM inside of biofilms, improved the penetration and permeabilization of the bacterial membrane.…”

“…Consequently, it is urgent to develop alternative antibacterial ways for the treatment of drug-resistant bacterial infections. [5][6][7][8][9][10][11][12] Saturated fatty acids are a class of promising biomaterials due to their low cost, biocompatibility, and biodegradability. 13,14 Among them, lauric acid (LA), a medium-chain fatty acid commonly found in coconut palm and milk, is reported to have selective bactericidal activity against Gram-positive bacteria by exerting a membrane-damaging effect.…”
