Skin rash by gefitinib is a sign of favorable outcomes for patients of advanced lung adenocarcinoma in Japanese patients

Abstract: Skin rash is one of the notorious adverse events of gefitinib as well as other epidermal growth factor receptor tyrosine kinase inhibitors. The differences of response rate and frequency of adverse events between ethnic groups are well known. Some reports demonstrated the correlation between development of rash and efficacy in Caucasian patients treated with erlotinib, gefitinib or cetuximab. We analyzed clinical course of Japanese patients of lung adenocarcinoma in order to assess the relation between adverse… Show more

“…The occurrence of skin rash is a predictor of gefitinib efficacy in patients with advanced NSCLC, as confirmed in numerous studies . Although this may be immune mediated, a PK relationship is still likely based on biology similar to that of other TKIs.…”

“…Therefore, we replaced these missing values by single imputation using the Expectation Maximization algorithm. This is an efficient way of handling missing data when it is missing at random or completely at random (Dong and Peng 2013).…”

“…Because the maximum amount of missing data for any given variable in our sample less than 7%, shown in Table 2, we judged 10 imputation sets more than sufficient for convergence. Further evidence of convergence can be determined by examining the worst linear function (WLF), a constructed statistic that converges more slowly than all other parameters in the model (Dong & Peng, 2013). Figure 1 shows no trend in the WLF, indicating that all the parameters have converged in the model.…”
