Small molecule antagonism of oxysterol‐induced Epstein–Barr virus induced gene 2 (EBI2) activation

Abstract: The Epstein–Barr virus induced gene 2 (EBI2) was recently identified as the first oxysterol-activated 7TM receptor. EBI2 is essential for B cell trafficking within lymphoid tissues and thus the humoral immune response in general. Here we characterize the antagonism of the non-peptide molecule GSK682753A, which blocks oxysterol-induced G-protein activation, β-arrestin recruitment and B-cell chemotaxis. We furthermore demonstrate that activation triggers pertussis toxin-sensitive MAP kinase phosphorylation, whic… Show more

“…EBI2 has been suggested previously to be coupled to the pERK 1/2 signaling pathway (Benned‐Jensen et al, ). To determine if activation of EBI2 with its endogenous agonist, 7 α 25HC, also leads to ERK phosphorylation in human astrocytes, from fetal cerebral cortex, the levels of phosphorylated ERK were examined.…”

“…Oxysterol‐signaling through various types of receptors is now known to be involved in regulating immune cell functions . In addition to its endogenous ligands, a number of small molecule ligands, both agonists and antagonists, have been characterized for EBI2 . These molecules are valuable tools to understand the functions of EBI2 and at the same time constitute potential drugs for diseases in which EBI2 may play a pathogenic role.…”

“…The authors concluded that the receptor has constitutive activity, although the possibility cannot be excluded that the signaling detected was due to low levels of oxysterols in the cell medium . Later, EBI2 was shown to also activate ERK and to recruit β‐arrestin . In addition, a study suggested that EBI2 increases the proliferation induced in B cells by B cell receptor cross‐linking, while another study saw no effect of 7α,25‐OHC treatment on LPS‐induced B cell proliferation .…”
