Small Untranslated RNA Antitoxin inBacillus subtilis

Abstract: Toxin-antitoxin (TA) modules are pairs of genes in which one member encodes a toxin that is neutralized or whose synthesis is prevented by the action of the product of the second gene, an antitoxin, which is either protein or RNA. We now report the identification of a TA module in the chromosome of Bacillus subtilis in which the antitoxin is an antisense RNA. The antitoxin, which is called RatA (for RNA antitoxin A), is a small (222 nucleotides), untranslated RNA that blocks the accumulation of the mRNA for a … Show more

“…The corresponding ORFs were expressed in E. coli under control of the inducible promoter P Bad . In contrast to TxpA from Bacillus subtilis which is not toxic in E. coli and does not induce lysis, 27 the expression of TxpA from E. faecalis V583 provoked cell death without lysis ( Fig. S2 and data not shown), showing the toxic effect of the protein.…”

“…5,[20][21][22][23] TA-I toxins, such as TxpA, are generally small hydrophobic proteins forming pores in membranes. 5,[24][25][26][27] Type II TA systems (TA-II) are usually made up by a bicistronic operon encoding the antitoxin protein that interacts with the toxin and inactivates its deleterious effects. Generally, TA-II toxin and antitoxin repress their own transcription via 'conditional cooperativity', a process relying on the DNA-binding properties of the antitoxin, the affinity between the toxin and the antitoxin, and their limited abundance.…”

“…The two respective RNA species overlap by 120 nucleotides and interact via a so-called kissing loop interaction. This interaction between txpA mRNA and the ratA RNA promotes degradation of the duplex in an RNase III-dependent manner (53,110,111). In the absence of ratA, the toxic TxpA peptide can be translated, which causes cell lysis.…”

“…Although the B. subtilis ymaH gene encodes a protein homologous to Hfq, this protein is not required for FsrAmediated inhibition of Sdh expression because a fur ymaH double mutant does not regain the ability to grow on succinate. To date, the function of Hfq remains unclear in B. subtilis and it is apparently not needed for the function of other known sRNAs (37,38). Perhaps because of the extensive sequence complementarity between FsrA and the sdhC mRNA (Fig.…”
