SMIT2 mediates all myo‐inositol uptake in apical membranes of rat small intestine

Abstract: This study presents the characterization of MI uptake in rat intestine using purified membrane preparations (BBMv). The two Na+‐coupled myo‐inositol (MI) cotransporters identified (SMIT1 and SMIT2) can be differentiated through inhibition studies using the selective substrates D‐chiro‐inositol (DCI, specific for SMIT2) and L‐fucose (specific for SMIT1). Results show that SMIT2 is exclusively responsible for apical MI transport in rat intestine. Other sugar transport systems present in apical membranes (SGLT1 a… Show more

“…11 Also, inositol acted as an osmolyte in some tissues, such as kidney medulla and brain, where osmolarity has a pivotal biological meaning. 12 In addition, inositol (either in the DCI or MI isoform) has been reported to mediate glucose uptake by improving insulin sensitivity. Insulin-mimetic effects of MI or its isomers seem to be derived from inositol phosphoglycan (IPG) which contains MI or DCI.…”

“…3k, l, o-q). This could be explained by the increased levels of fumarate and myo-inositol in cultivated lettuce compared to wild accessions 24,26 . The fumarate derivative, dimethyl-fumarate (DMF), has anti-inflammatory and antioxidant capacity and it can also improve the tight-junction protein expression 27 .…”

“…p = 0.0008 for rs28540434). SLC5A11 is expressed in the gastrointestinal tract and kidney and has been found to be responsible for apical MI transport in the rat intestine (48,49). Hence, variation in expression or function of this gene represents a plausible mechanism by which plasma MI could be modulated.…”
