Smoking and Pancreatic Cancer: Smoking Patterns, Tobacco Type, and Dose-Response Relationship

Abstract: Pancreatic cancer (PC) is the primary cause of cancer death in the United States and Europe. Despite remarkable advances in the molecular understanding of PC and advances in new therapeutic approaches, PC remains a disease with a poor prognosis. Although evidence indicates that long-term smoking is a major cause of PC, the molecular pathways behind smoking-induced PC pathogenesis are not fully understood. Smoking cessation can significantly reduce the occurrence of PC. This review explores the proce… Show more

“…In addition, the number of years smoking was negatively correlated with the survival rate of pancreatic cancer patients [20]. Of the 7,000 chemicals found in tobacco smoke, at least 250 are harmful, and 60 are carcinogenic [5]. Cigarette smoke promotes the metastasis of pancreatic cancer by upregulating the expression of MUC4 in pancreatic cancer tissue [21].…”

“…The modifiable risk factors for pancreatic cancer include behavioral factors and metabolic factors. Cigarette smoking is a well-established behavioral risk factor for various cancers, such as tracheal, bronchus and lung cancer, laryngeal cancer, head and neck squamous cell carcinomas, esophageal cancer, colorectal cancer and pancreatic cancer [2][3][4][5][6][7]. Smoking was the second leading risk factor for mortality in both sexes in 2015 [8].…”

“…Our findings on smoking and PC risk are consistent with previous population-based studies showing that current and passive smoking were associated with an increased risk of PC. 9,[18][19][20][21][22] A recent metaanalysis on the association between smoking and PC risk found a higher risk for current smokers than non-smokers. 23 Similarly, largescale studies from Europe 24 and Asia 25 showed that smoking is associated with a 2-fold increased risk of PC.…”

“…Among them, tobacco use is, to the present knowledge, the most important one. Indeed, a recent review by Subhan et al 9 . showed that smoking is a strong risk factor for PC, leading to progressive fibrosis of the pancreatic glandular tissue and stroma.…”
