Sodium and chloride co‐transport by preimplantation rabbit blastocysts.

Abstract: SUMMARY1. Influxes of Na+ and C1-across the trophectodermal epithelium of 6 day post coitum (p.c.) rabbit blastocysts are partially interrelated. This relationship was investigated under conditions in which either the external [Na+] or [Cl-] concentration was varied. Our results indicate that an ouabain-insensitive, furosemide-sensitive, Na+: Cl-co-transport system exists in the 6 and 6'5 day p.c. rabbit embryo, but not in the 5 day p.c. blastocyst.2. There was no dependence of NaCl uptake on the presence of… Show more

“…4), as a result of transtrophectodermal C efflux down the imposed C1-gradient and the resulting unquenching of the dye. This was also seen in the presence of SO 4 2 -as substitute anion. It was not possible to study both C1-exit and C1-re-entry without compromising the integrity of the blastocyst; therefore this C1-efflux was used to study the mechanism and regulation of transtrophectodermal Cl-transport.…”

“…In the rabbit, 36 C1-uptake is unaffected by DIDS, but a furosemide-sensitive Na+/C1 -co-transporter is present in Day 6 blastocysts, although not in earlier ones [4]. Thus the rat blastocyst appears to share with the rabbit, but not the mouse, a transcellular pathway for Cl-transport.…”

“…Cyclic AMP decreased the active transport of Na+ and Cl~in the blastocyst (Benos & Biggers, 1983). To determine whether a decrease in ion transport affected glucose metabolism to C02, we tested the effect of chlorophenylthio cyclic AMP (CPT cAMP; 10~4m) on C02 production.…”
