Sodium Cromoglycate Prevents Exacerbation of IgE-Mediated Food-Allergic Reaction Induced by Aspirin in a Rat Model of Egg Allergy

Abstract: Background: Aspirin (ASP)-facilitated absorption of ingested allergens is considered an exacerbating factor in the development of food allergy. Sodium cromoglycate (SCG) is used for the treatment of atopic dermatitis with food allergy, but the efficacy of SCG in ASP-exacerbated food-allergy reactions is unclear. In this study, we evaluated the effect of SCG on ASP-exacerbated food-allergic reactions, as well as allergen absorption, in egg-allergic model rats. Methods: Plasma concentrations of ovalbumin (OVA) a… Show more

“…Previous reports have shown that aspirin induced and exacerbated allergic symptoms by promoting absorption of ingested allergens in rats and patients with IgE-mediated food allergies [79,11]. However, there are no reports regarding the effect of aspirin on the sensitization to ingested allergens.…”

“…We previously demonstrated that OVA were absorbed from intestine as an intact form in the presence of protease inhibitors using in situ intestinal perfusion technique [5]. In addition, aspirin elicited and/or exacerbated the allergic symptoms by increasing oral absorptions of OVA [9]. These reports suggested that aspirin could enhance the oral absorption of OVA as an intact form at least partly.…”

“…In this study, OVA, ω5-gliadin, Hokushin gluten or 1BS-18 gluten were intravenously administered because we can evaluate their allergenicities while excluding processes of digestion and absorption. Rectal temperature was measured using a specific rectal thermometer for rats (Shibaura Electronics, Saitama, Japan) before and 30 min after intravenous challenge according to a previous report [16]. After measuring the rectal temperature at 0 min, OVA, ω5-gliadin, Hokushin gluten or 1BS-18 gluten (10 mg/kg) were intravenously injected via a canula inserted in the femoral vein.…”
