Sodium pentobarbital dosages for exsanguination affect biochemical, molecular and histological measurements in rats

Abstract: Rodents are widely used for animal research in Egypt. Pentobarbital is the most common anesthetic agent; however overdoses may affect the experimental outcomes and limit the use of tissues. To investigate the effects of sodium pentobarbital overdoses during exsanguination, three groups (6 rats/ group) of male and female rats were injected i.p. with 50, 100 and 150 mg/kg of sodium pentobarbital, then carotid exsanguination was performed immediately after loss of consciousness. Hypoxiainducible factor 1-alpha (H… Show more

“…Sprague–Dawley rats, 150–200 gm, were given 4 × 3 cm 2 dorsal full-thickness excisions under anesthesia with 50 mg/kg intraperitoneal pentobarbital ( Mohamed et al, 2020 ). The procedures were approved by the Ethical Committee of Misr University for science and technology (reference number: PH 7) and followed the guidelines for the care and use of laboratory animals published by US NIH publication No.…”

“…Our study had several limitations, including the use of pentobarbital at such a high dose (150 mg/kg) during exsanguination. This dose ‎might have influenced some cytokines and biochemical parameters, such as liver ‎enzymes, urea ‎, and creatinine [ 62 ].…”

“…; Merk, Germany; Cat No: #P0225) and then immediately placed on a heating pad (37°C) to minimize animal suffering. Sodium pentobarbital is the most common anesthetic agent in animal experiments and has the following advantages: simple administration, fast-action, good anesthetic effect and longer lasting, as described previously (Mohamed et al, 2020;Olsen & Li, 2011). After anesthetization, the rat was quickly decapitated, and the brain tissue was removed and immersed in frozen artificial cerebrospinal fluid (ACSF) comprising (in mM per 1,000 ml) 119 NaCl (CAS: 7647-14-5), 3.5 KCl (CAS: 7447-40-7), 1.3 MgSO 4 (CAS: 7487-88-9), 2.5 CaCl 2 (CAS: 10043-52-4), 1 NaH 2 PO 4 (CAS: 13472-35-0), 26.2 NaHCO 3 (CAS: 144-55-8), and 10 glucose (CAS: 50-99-7), pH 7.4.…”
