Sodium Valproate‐lnduced Cardiovascular Abnormalities in the JchlCR Mouse Fetus

Abstract: Sodium valproate was administered to Jc1:ICR mice in order to evaluate teratogenicity in the cardiovascular system. A single dose of 600 mg/kg of sodium valproate was injected intraperitoneally on gestational day 6 , 7, 8 or 9. On day 18 of gestation, dams were laparotomized to observe incidence and type of cardiovascular abnormality in live fetuses. Cardiovascular abnormalities were found most frequently in the group treated on day 7 , being recognized in 86% of litters (19/22) and in 29% of live fetuses (70/… Show more

“…One of the reasons that so many theories and hypotheses have been proposed is the absence of an adequate animal model for experimental analysis of the pathogenesis of TGA (Pexider, 1992). Various teratogens that can be used to generate this anomaly have been reported, such as trypan blue (Monie, 19661, hyperbaric oxygen (Miller, 1971), neutron irradiation (Okamoto, 1976;Asami and Koizumi, 1984), and sodium valproate (Sonoda, 1991). Recently, we established a mouse model of this heart malformation in which TGA is found in 80% of fetuses derived from dams treated with a single dose of all-trans retinoic acid (RA) (Irie, 1990;Yasui, 1995).…”

“…1 At the same time its exposure during pregnancy has also been found to cause various types of birth defects including craniofacial defects, cle palates, spina bida and cardiovascular and limb related anomalies, 2-4 which have been collectively termed as 'Fetal Valproate Syndrome'. [6][7][8][9][10][11][12][13] As more than 50% of pregnancies are unplanned, sometimes the situation becomes critical when a woman nds out that she has to undergo treatment with VPA while she is pregnant. “…5 A similar pattern of embryonic/foetal malformations has also been demonstrated among various experimental species. [6][7][8][9][10][11][12][13] As more than 50% of pregnancies are unplanned, sometimes the situation becomes critical when a woman nds out that she has to undergo treatment with VPA while she is pregnant. In a few cases it becomes more crucial when they can neither go for a Medical Termination of Pregnancy (MTP) nor keep the treatment pending until delivery.…”
