Solid medium for the direct isolation of bacterial colonies growing with polycyclic aromatic hydrocarbons or 2,4,6-trinitrotoluene (TNT)

Abstract: Isolation of hydrocarbon-degrading bacteria is a key step for the study of microbiological diversity, metabolic pathways and bioremediation, however current methods lack simplicity and versatility. We developed an easy method that allows the screening and isolation of bacterial colonies capable of degrading hydrocarbons, such as diesel or polycyclic aromatic hydrocarbons (PAHs), as well as the pollutant explosive, 2, 4, 6-trinitrotoluene (TNT). The method uses a two-layer solid medium, with a layer of M9 mediu… Show more

“…These isolated species were previously reported as cold-tolerant species 41 , 43 45 . Several species, including P. silesiensis 12 and P. frederiksbergensis JAJ28 T 45 , have been reported to be capable of growing and degrading phenanthrene. However, to our knowledge, phenanthrene degradation by P. fildesensis , Paeniglutamicibacter terrestris and Pseudarthrobacter humi at low temperatures has never been studied.…”

“…Therefore, the identification of indigenous Antarctic microorganisms with PAH-degrading capabilities is essential for the bioremediation of contaminated Antarctic environments. To date, various PAH-degrading bacteria have been isolated from Antarctica, including Pseudomonas , Rhodococcus , Sphingobium , and Acinetobacter 12 , 13 , which are common genera found in soils from both polar and temperate regions. Dietz-Vargas et al 12 isolated Acinetobacter sp.…”

“…Genomic DNA extraction was carried out with the Wizard Genomic DNA Purification Kit (Promega) following the manufacturer’s instructions, and the amplification of a nearly full-length 16S rRNA gene fragment was performed as previously described [18]. Resulting amplicons were sequenced at AUSTRAL-omics (Chile).…”

“…The need to restore aquatic ecosystems dictates the search for efficient, cost-effective and environmentally friendly methods for the elimination of explosives, which corresponds to the processes of biological treatment (Lewis et al, 2004;Shemer et al, 2018;Dietz-Vargas et al, 2023). Serrano-González et al (2018) have proposed combining several kinds of biological TNT treatment: anaerobic, aerobic, and enzymatic processes.…”
