Solo and Keratin Filaments Regulate Epithelial Tubule Morphology

Abstract: Epithelial tubules, consisting of the epithelial cell sheet with a central lumen, are the basic structure of many organs. Mechanical forces play an important role in epithelial tubulogenesis; however, little is known about the mechanisms controlling the mechanical forces during epithelial tubule morphogenesis. Solo (also known as ARHGEF40) is a RhoA-targeting guanine-nucleotide exchange factor that is involved in mechanical force-induced RhoA activation and stress fiber formation. Solo binds to keratin-8/kerat… Show more

“…Its high expression is found in mammalian single-layered epithelia, such as single columnar epithelium lining the oviduct. Its main role, together with keratin 8, is the formation of intermediate filaments (IF), creating structural framework of epithelial cells [33]. Although keratins are mainly known for their ability to strengthen and stabilize cells, they can indirectly influence intercellular adhesion.…”

“…AGAP3 is a member of a family of proteins containing functional ArfGAP domains and GTpase-like domains (AGAP1-3) with bifunctional enzyme activity ( Oku and Huganir, 2013 ). Solo (arghef40) is a guanine nucleotide exchange factor that targets RhoA and regulates cell morphology by interacting with keratin filaments ( Nishimura et al, 2018 ). DOC-2 / DAB-2 interacting protein (Dab2IP) is a member of the Ras gtpase activating protein (GAP) family ( Lee et al, 2012 ; Qiao et al, 2013 ).…”

“…The metastasome signature comprises seven genes, three of which overlap with the fibrosome. The other four genes are involved in the regulation of cell movement [55-57] or in migration and structural remodeling [58]. We identified upregulation of these genes in PD.…”
