Soluble E‐selectin, other markers of inflammation and disease severity in children with atopic dermatitis

Abstract: E-selectin, P-selectin, intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) are membrane-bound adhesion molecules which mediate the attachment of leucocytes to endothelial cells. These molecules are preferentially expressed on activated endothelium. The soluble forms of these molecules (sE-selectin, sP-selectin, sICAM-1 and sVCAM-1) are present in the circulation as a result of shedding. Some of the soluble adhesion molecules have been thought to reflect disease activity i… Show more

“…The plasma concentration of soluble E-selectin in children significantly correlated with disease activity, 17 whereas soluble P-selectin, soluble ICAM-1, and soluble VCAM-1 levels were unchanged. Soluble E-selectin appears to be the best serologic marker to determine disease activity in AD thus far.…”

“…Next, the results were translated into arbitrary units (AU) by relating the obtained oxLDLÀLDL (OD) values of the test samples to the oxLDLÀLDL (OD) value of the reference in the same ELISA-plate. Levels of sICAM-1 were measured by capture ELISA (HBT, Uden, The Netherlands) using a monoclonal antibody to sICAM-1 (HM.2) as capture antibody and a biotin-labelled anti-ICAM-1 as detection antibody, as described elsewhere (Wolkerstorfer et al, 1998).…”

“…31 The index has shown agreement with global assessments of disease severity 35 as well as with various circulatory factors thought to reflect disease activity in atopic dermatitis. [32][33][34] However, problems with interobserver variation have occurred. In the original description of the scoring system, reliability was tested using 10 trained observers and 10 slides.…”
