Somatostatin Acts by Inhibiting the Cyclic 3′,5′-Adenosine Monophosphate (cAMP)/Protein Kinase A Pathway, cAMP Response Element-Binding Protein (CREB) Phosphorylation, and CREB Transcription Potency

Abstract: Somatostatin (SRIF) was discovered as an inhibitor of GH secretion from pituitary somatotroph cells. SRIF analogs are very effective agents used to treat neuroendocrine tumors and are now being used with increasing frequency in clinical trials to treat more aggressive malignancies. However, the cellular components mediating SRIF signal transduction remain largely unknown. We have stably overexpressed the SRIF type 2 receptor (SST2) in GH4 rat somatomammotroph cells, establishing a physiologically relevant mode… Show more

“…In GH 4 C 1 cells, SRIF-action is mediated specifically via Ga i2 (Liu et al 1994), as PTX suppresses SRIF-mediated inhibition of VIPinduced cAMP (Koch et al 1985). PTX treatment similarly attenuated SRIF action in GH 4 cells overexpressing sst2 (Tentler et al 1997). In addition, PTX attenuated SRIFdependent inhibition of GHRH-induced GH in MtT/SGL somatotroph cells (Morishita et al 2003).…”

“…sst1, sst2, sst3, and sst5 all mediate SRIF inhibition of adenylate cyclase in pituitary cells (Tentler et al 1997, Cervia et al 2003, Ben-Shlomo et al 2005. SRIF inhibited forskolin-induced cAMP production, PKA activation, CREB phosphorylation, and transcription potency, while overexpression of the PKA catalytic subunit suppressed SRIF action in sst2 stable transfectant GH 4 cells (Tentler et al 1997).…”

“…The cellular actions of SRIF are complex, but a major feature of SRIF receptor activation is inhibition of cAMP and Ca 2ϩ currents (Meriney et al, 1994;Tentler et al, 1997) and opening of an inwardly rectifying K ϩ channel (Inoue et al, 1988). In relation to DRG cells, SRIF has been shown to decrease Ca 2ϩ current conductance (Gammon et al, 1990;Taddese et al, 1995).…”

“…The somatotroph cells located in anterior pituitary gland secretes GH under the influence of growth hormone releasing hormone (GHRH) and ghrelin, whereas somatostatin inhibits GH secretion [50][51][52][53]. Ghrelin is an endogenous ligand for the GH secretagogue receptor (GHS-R).…”
