Sophoranone, extracted from a traditional Chinese medicine Shan Dou Gen, induces apoptosis in human leukemia U937 cells via formation of reactive oxygen species and opening of mitochondrial permeability transition pores

Abstract: Screening of various natural products in a search for novel inducers of apoptosis in human leukemia cells led us to identify the strong apoptosis-inducing activity in a fraction extracted with methanol from the roots of Sophora subprostrata Chun et T. Chen. We purified the compound that induced apoptosis in human leukemia cells and identified it as sophoranone. Sophoranone inhibited cell growth and induced apoptosis in various lines of cells from human solid tumors, with 50% inhibition of growth of human stoma… Show more

“…Thus the cytotoxicity of flavonoids toward malignant cells has been examined. [28][29][30][31][32][33][34][35] On the other hand, there are many reports that flavonoids protect various cell types from oxida- tive stress. 11,12,36) Since oxidative stress is suggested to cause many diseases including cardiovascular and neurodegenerative diseases, 37) flavonoids are possible dietary preventives against these diseases.…”

“…Phytochemical investigations have revealed that the plant accumulated isoprenyl-substituted flavonoids and lupin alkaloids as its main constituents (Ding & Chen, 2006. Pharmacological studies showed that the isoprenylated flavanones isolated from this species could inhibit cell growth and induce apoptosis on various cell lines from human solid tumors and in human leukemia U937 cells (Kajimoto et al, 2002). Sophora moorcroftiana is an endemic shrub species in Tibet, China, and is mainly distributed in the wide valleys and the middle reaches of several main tributaries of Yalu Tsangbo River (Nianchu and Lhasa Rivers).…”
