Spatial control of nucleoporin condensation by fragile X‐related proteins

Abstract: Nucleoporins (Nups) build highly organized nuclear pore complexes (NPCs) at the nuclear envelope (NE). Several Nups assemble into a sieve-like hydrogel within the central channel of the NPCs. In the cytoplasm, the soluble Nups exist, but how their assembly is restricted to the NE is currently unknown. Here, we show that fragile X-related protein 1 (FXR1) can interact with several Nups and facilitate their localization to the NE during interphase through a microtubule-dependent mechanism. Downregulation of FXR1… Show more

“…We also observed cytoplasmic inclusions of nuclear pore components, as well as of RanGAP1,a key protein regulating nucleocytoplasmic shuttling function, which recapitulates previous observations in ALS patients 15,21 . These cytoplasmic nuclear pore inclusions were analogous to the inclusions recently observed upon FXR1 downregulation 74 . Hence, further research is needed to determine their origin and their potential relevance to ALS, especially as these cytoplasmic nucleoporin inclusions co-occur with p62 inclusions also found in ALS patients, in particular C9ORF72 HRE patients [75][76][77] .…”

“…Membrane-free cytoplasmic Nups are also described as largely soluble [ 84 ] or as condensates in stress granules during a variety of insults [ 85 ]. Very recently, an additional form of cytoplasmic Nup congregation with liquid–liquid interaction properties was found in the absence of the fragile X-related protein [ 86 ]. Yet, none of these structures are reminiscent of, and probably do not relate to, LD.…”

“…Different types of fragile X‐granules are expressed in neurons, where they regulate transport, localization, stability, and translation of target RNAs. In the current study, Agote‐Aran et al () expand our view on FXR proteins and show that they also control the assembly of granules containing nucleoporins, the building blocks of nuclear pore complexes (NPCs).…”
