Spatiotemporal immune zonation of the human kidney

Abstract: Tissue-resident immune cells are important for organ homeostasis and defense. The epithelium may contribute to these functions directly or by cross-talk with immune cells. We used single-cell RNA sequencing to resolve the spatiotemporal immune topology of the human kidney. We reveal anatomically defined expression patterns of immune genes within the epithelial compartment, with antimicrobial peptide transcripts evident in pelvic epithelium in the mature, but not fetal, kidney. A network of tissue-resident myel… Show more

“…Major cell types and subclusters were identified based on cell type-specific markers ( Fig. S2C, D, Dataset S1) (8)(9)(10). Each nephron segment performs unique reabsorptive and secretory functions to transform filtrate into urine, and this is reflected by segment-specific expression of all detected solute linked carriers, ATPases and channels ( Fig.…”

“…Recent scRNA-seq analyses have revealed unexpected stromal heterogeneity in both developing and adult kidney (8,9,23). With Harmony integration, we combined the stromal clusters from all timepoints to identify eight stromal cell subclusters (Fig.…”

“…We also compared the immune compartment of heart, skeletal muscle and adult kidney 79 by analyzing 40 206 cells and nuclei. Across the three tissues there was extensive overlap of lymphoid immune populations (B cells, CD4+ and CD8+ T cells, and NK/NKT cells).…”
