Metabarcoding Analysis of Ichthyoplankton in the East/Japan Sea Using the Novel Fish-Specific Universal Primer Set

The spatiotemporal distribution of fish larvae and eggs is fundamental for their reproduction and recruitment in aquatic ecosystems. Here, a metabarcoding strategy was employed as an alternative to a conventional ichthyoplankton survey, which requires a considerable amount of time, labor, and cost. First, a piscine-specific universal primer set (FishU) was designed to amplify the region, flanking the highly conserved mitochondrial 12S and 16S ribosomal genes, and it was optimized for the MiSeq platform. Based on both in silico and in vitro analyses, the newly designed FishU primers outperformed the two previously reported fish-specific universal primer sets (ecoPrimer and MiFish) in taxon coverage, specificity, and accuracy in species identification. The metabarcoding results by FishU primers successfully presented the diversity of ichthyoplankton directly from the zooplankton net samples in the East/Japan Sea, presenting more accurate and plentiful species numbers than those by MiFish primers. Thus, the metabarcoding analysis of ichtyoplankton using the newly designed FishU primers is a promising tool for obtaining useful data to understand the reproduction of fish, such as spawning sites, reproductive periods, population structures, feeding ecology, and diet.

The aim of this study was to assess the temporal variability in the trophic structure of a fish community and the trophic levels of dominant fish species off the eastern coast of Korea. We analyzed the community composition and the stable carbon and nitrogen isotope ratios (δ 13 C and δ 15 N) for fish species in Hupo coastal waters over four seasons. Fish assemblages were mainly dominated by resident benthic and benthopelagic fish. In contrast, there were low numbers of pelagic fish, including migrating and occasional species, which varied significantly by season. The fish assemblages generally had relatively wide ranges of δ 13 C and δ 15 N (− 21.1 to − 16.3‰ and 10.0 to 14.2‰, respectively), reflecting a clear distinction between resident (mostly benthic or benthopelagic) vs migratory (mostly pelagic) fish. Resident species showed no seasonal differences in their isotopic signatures, whereas pelagic species showed temporal variability in δ 13 C and δ 15 N values. The isotopic niche of fish assemblages showed distinctly different patterns between spring-summer and winter-fall with the seasonal changes in the fish community. These patterns might be associated with seasonal variation in the fish community caused by seasonal changes in environmental conditions (e.g., hydrographic parameters and food availability). Our results can offer baseline information/trophic data for ecosystem models that can be used to evaluate the effects of climate on marine ecosystems.

Antifreeze Activity in Temperate Fish from the East Sea, Korea

Antifreeze proteins and glycoproteins [AF(G)Ps] constitute a group of proteins that lower the freezing but not the melting points of aqueous solutions, enabling polar and north-temperate fish to survive in ice-laden environments. However, little is known about antifreeze activity in temperate fish; such work would extend our knowledge on the functions and evolution of AF(G)Ps. In the present study, we screened for antifreeze activity in temperate fish caught off the coast of Jumunjin (37.89°N), Gangneung, Korea. Thermal hysteresis (TH) and the ability to inhibit ice recrystallization (IR) in blood, liver, and muscle samples from nine fish were examined to assess antifreeze activity. As the East Sea off the coast of Jumunjin is ice-free year round, we thought it most unlikely that the fish would express antifreeze proteins. Surprisingly, the blood of Pleurogrammus azonus and three types of tissue from Gymnocanthus herzensteini, Zoarces gilli and Kareius bicoloratus exhibited measurable TH values together with the ability to trigger characteristic morphological changes in ice crystals. Blood samples from the three species also evidenced ice recrystallization (IR) inhibition. This implies that AF(G)Ps or other antifreeze-like substances are present in temperate fish even under nonfreezing conditions. These results contribute to our understanding of the functions and origins of antifreeze activity in fish.
