Specific cleavage of ribosomal RNA and mRNA during victorin‐induced apoptotic cell death in oat

Abstract: SummaryHere we report that rRNA and mRNA are specifically degraded in oat (Avena sativa L.) cells during apoptotic cell death induced by victorin, a host-selective toxin produced by Cochliobolus victoriae. Northern analysis indicated that rRNA species from the cytosol, mitochondria and chloroplasts were all degraded via specific degradation intermediates during victorin-induced apoptotic cell death but, in contrast, they were randomly digested in necrotic cell death induced by 30 mM CuSO 4 and heat shock. This… Show more

“…Circles denote total germination as in b. Asterisks in b and d indicate differences between non-aged control seeds and Aged or dead seeds at a level of P \ 0.01 (one-way ANOVA) (Bray and Chow 1976;Boubriak et al 2000;Brockelhurst and Fraser 1980;Cheah and Osborne 1978;Osborne 2000). The final, or execution, phase of PCD is characterised by inter-nucelosomal DNA fragmentation (Elbaz et al 2002;Hengartner 2000;Thomas and Franklin-Tong 2004) and can include RNA degradation (Hoat et al 2006;Perrin et al 2004). It has been reported occasionally for apoptotic animal cells that rRNA remained intact (Houge and Doskeland 1996) and conversely, rRNA fragmentation occurred without DNA laddering (Samali et al 1997).…”

“…ZnCl 2 functions as a Ca 2+ antagonist preventing calcium metabolism, and inhibits victorin-induced apoptotic cell death in oat (Hoat et al 2006). To examine the effect of ZnCl 2 on necrotic cell death in wheat hybrids, we treated seedlings showing the three types of hybrid necrosis with ZnCl 2 .…”

“…In plants, ribosomal RNA is specifically cleaved during apoptotic cell death induced by victorin in oat cells [10]. However, rRNA cleavage does not appear as a general feature associated with apoptosis [21].…”

“…Under these conditions, alamethicin induced specific rRNA cleavage. Interestingly, rRNA cleavage has previously been shown to occur in plant and animal cell lines during apoptosis [10] [11]. Apoptosis is a form of programmed cell death (PCD) 2 ) that plays fundamental roles in tissue homeostasis and stress responses [12].…”
