Specificity of MicroRNA Detection on a Power-free Microfluidic Chip with Laminar Flow-assisted Dendritic Amplification

Abstract: MicroRNAs (miRNAs) are attracting considerable attention as potential biomarkers for the early diagnosis of cancer. We have been developing a detection method for miRNAs on a microfluidic chip with external-power-free fluid pumping and enzyme-free amplification. The assay is completed within 20 min. Here, we describe the specificity of this miRNA detection method. First, the specificity against mismatched sequences was investigated. The nonspecific detection of a 2-nucleotide mismatched sequence was negligible… Show more

“…Therefore, a device in which sample-preparation steps (e.g., enzymatic steps for PCR-based amplifications) are removed, would represent an improvement in microRNAs detection and quantification. Some tentative approaches are under study like the integration of a dynamic chemistry for “Single Nucleobase Labeling” with a bead-based platform (Luminex®) (Venkateswaran et al, 2016 ), or the use of a power-free microfluidic chip involving a technology based on laminar flow–assisted dendritic amplification (LFDA) (Hasegawa et al, 2017 ). In the former case the same technology was used to detect microRNAs from serum samples (Rissin et al, 2017 ).…”

“…The specificity of miRNA detection on the power-free microfluidic chip was further studied under various conditions 33 . First, using a human miR-204 detection system, non-specific responses from mouse miR-211 (1-base mismatch) and human miR-211 (2-base mismatch) were examined.…”

“…1 piRNAs are single-stranded RNAs of 26 -31 nucleotide (nt) length, which are slightly longer than the better-known microRNAs (21 -25nt). 2,3 Their 3′-terminal nucleotides feature 2′-O-ribose methylation by methyltransferase HEN1, which can enhance the piRNA stability. 4,5 The binding of piRNAs to Piwiclass Argonaute proteins 6 leads to a silencing of the target complementary messenger RNAs.…”
