Specificity protein-1 and -3 trans-activate the ovine placental lactogen gene promoter

Abstract: The proximal promoter (-383/+16) of the ovine placental lactogen (oPL) gene provides trophoblast-specific expression in vitro. Footprint 6 (FP6; -319/-349) lies within this region, and transfection of two-base pair mutations across FP6 into BeWo cells identified potential binding sites for CCAAT-enhancer binding protein (CEBP) and Specificity Proteins (Sp). Transfection of CEBP dominant negative or over-expression constructs did not impact transactivation of the proximal promoter. However, Sp1 and Sp3 over-exp… Show more

“…CSH1 expression is well correlated to the maturation of BNCs among bovine trophoblasts (Nakano et al 2005); however, information on the regulation and transcription factors for CSH1 is still limited. Some elements, AP2, GATAs, PURA, and SPs, have been reported as activation factors for this gene, and CSH1 expression was closely related to the implantation process spatiotemporally (Limesand et al 2004, Jeckel et al 2009). GATA may play a regulatory role of CSH1 expression and differentiation of BNCs (Bai et al 2011).…”

“…For cold competition (CC), 200-fold molar excess of unlabeled homologous oligonucleotides were added to EMSA binding reactions. As described earlier [ 63 ], for super-shift assays, 1 μl of normal rabbit serum or HIF-1α antiserum (Active Motif) was pre-incubated with NE in the 1x binding buffer for 2 hours at 4°C. This was followed by addition of biotinylated probe and poly (dI-dC).…”

“…The positive effect of leptin on CSH2 expression also may encourage the continued use of CT1 cells to study CSH2 gene expression. Previous examinations of bovine or ovine CSH2 expression regulation required heterologous transfection studies using human and rodent choriocarcinoma lines [18,19]. Marginal CSH2 expression occurs in bovine trophoblast cell lines, including CT1 cells under normal growth conditions.…”
