Spectral sensitivity of guppy visual pigments reconstituted in vitro to resolve association of opsins with cone cell types

Abstract: The guppy (Poecilia reticulata) shows remarkable variation of photoreceptor cells in the retina, especially those sensitive to middle-to-long wavelengths of light. Microspectrophotometry (MSP) has revealed varying "green", "green-yellow" and "yellow" cone cells among guppies in Trinidad and Venezuela (Cumana). In the guppy genome, there are four "long-wave" opsin loci (LWS-1, -2, -3 and -4). Two LWS-1 alleles have potentially differing spectral sensitivity (LWS-1/180Ser and LWS-1/180Ala). In addition, two "mid… Show more

“…In bony fishes, the opsin repertoire differs among species (Gojobori & Innan, 2009; Yokoyama, 2000). Generally, the surface‐dwelling species, which habit the luminous superficial area of water, have a large cone opsin repertoire, as seen in zebrafish ( Danio rerio ) (Chinen, Hamaoka, Yamada, & Kawamura, 2003), medaka ( Oryzias latipes ) (Matsumoto, Fukamachi, Mitani, & Kawamura, 2006), and guppy ( Poecilia reticulata ) (Kawamura et al., 2016). Conversely, dark deep‐sea fish tend to lose cone cells and corresponding opsins, whereas they possess a high proportion of rod cells, which contribute to dim light vision as in the case of lizardfish (Hope, Partridge, Dulai, & Hunt, 1997).…”

In addition, we also calculated absolute cone stimulation (ACS) among the different light environments, using both (i) the measured peak absorbance (λ max ) of their respective photoreceptor MSP values (; and references therein) and (ii) Kawamura et al.’s () measured peak absorbance (λ max ) values for each opsin visual pigment reconstituted in vitro ( and ).…”
