Spectrophotometric determination of total protein in serum using a novel near-infrared cyanine dye, 5,5′-dicarboxy-1,1′-disulfobutyl-3,3,3′,3′-tetramethylindotricarbocyanine

Abstract: The application of near-infrared (NIR) dyes (lambda (em) > 750 nm) to the analysis of biological samples shows much promise, because the long emission wavelengths of such dyes allow interferences from biomolecule matrices to be minimized. In this paper, a novel NIR dye, 5,5'-dicarboxy-1,1'-disulfobutyl-3,3,3',3'-tetramethylindotricarbocyanine (DCDSTCY) has been developed for the spectrophotometric determination of total protein in serum. Under acidic conditions, the binding of DCDSTCY to proteins caused a new … Show more

“…7 Once binding with the biomolecules, the fluorescence quantum yield of the dyes was enhanced a lot because of the rotation hindrance. 8 This is very advantageous for utilizing them as non-covalent labels 9,10 because the free form of the dye has a much lower quantum yield than the binding one. The cyanine dyes have also found numerous applications as photosensitizers and nonlinear optical materials.…”

“…In this work, we reported a novel NIRF/MR bimodal imaging probe, Gd-NMC-3 (Scheme ), composed of three fragments: (1) a near-infrared fluorophore, ICG derivative DCDSTCY, as a core group can not only act as a NIRF portion but also show high selectivity to the tumor; , (2) a MR imaging signal unit, named Gd-DOTA, could provide the anatomic information with unlimited tissue penetration depth; and (3) the linker butane diamine could covalently connect with the NIRF component and MR component by amide bonds. As confirmed by the cellular uptake experiment, the probe could be internalized into cancer cells by OATPs and NTCP, indicating that Gd-NMC-3 has an excellent specificity to tumor tissues.…”

“…In this work, we reported a novel NIRF/MR bimodal imaging probe, Gd-NMC-3 (Scheme 1), composed of three fragments: (1) a near-infrared fluorophore, ICG derivative DCDSTCY, 44 as a core group can not only act as a NIRF portion but also show high selectivity to the tumor; 33,45 and reasonable relaxation property in tumor sites, which demonstrated that Gd-NMC-3 is a superior NIRF/MR bimodal imaging probe. On the basis of the perfect delineation of the liver tumor, Gd-NMC-3 was applied to guide the surgical resection of orthotopic and subcutaneous xenograft tumors in mice under intraoperative conditions.…”

“…It is defined at a wavelength of 780 ~ 2 526nm spectral region by the Material Measuring Association of the USA. The near-infrared spectroscopy analysis technique, which has a series of advantages: in vivo analysis, rapid measurement, simple operation and low measuring cost, is widely used in various material properties measurement in recent years [11][12][13]. Alcohol has obvious transmission phenomenon in the near infrared region, and its concentration is positively correlated with transmittance rate.…”
